Bestimmung der Phenolcarbonsäuren (Hydroxybenzoesäuren und Hydroxyzimtsäuren) in Rebblättern, Weintrauben und Wein mittels Mikro-Polyamid-Dünnschichtchromatographie

Zur schnellen zweidimensionalen Trennung der Hydroxybenzoesäuren und Hydroxyzimtsäuren auf Mikro-Polyamid-Fertigplatten wurde eine Reihe von Fließmittelkombinationen untersucht. Die am besten geeigneten Fließmittel werden tabellarisch zusammengefaßt. In wäßrigen Fließmitteln lassen sich die cis- und transisomeren Zimtsäureabkömmlinge gut voneinander trennen. Zur Abtrennung der Methoxyverbindungen von den entsprechenden Hydroxyverbindungen hat sich ein Fließmittelgemisch aus Benzol-Eisessig sehr gut bewährt. Die Laufzeiten betragen je nach Fließmittel 20 bis 80 Minuten.
Es wird ein Überblick über die in Trauben und Rebblättern vorhandenen Phenolcarbonsäuren gegeben. Salicylsäure und Gentisinsäure treten in freier Form in den Trauben erst in späteren Reifestadien auf.
Die Hauptunterschiede im Phenolcarbonsäuren-Muster der untersuchten Sorten sind durch quantitative Differenzen von Syringasäure, Ferulasäure und zwei noch nicht identifizierten Komponenten bedingt.
Analysis of the phenolcarboxylic acids (hydroxybenzoic acids and hydroxycinnamic acids) in grape leaves, berries, and wine by micro-polyamide thin-layer chromatography
In order to achieve a rapid two-dimensional separation of hydroxybenzoic acids and hydroxycinnamic acids on micro-polyamide layer sheets, a series of solvent combinations was investigated. The most suitable solvents are giveri in Table 1. In a watery solvent, the cis-isomer and trans-isomer cinnamic acid derivatives can easily be separated from each other. In order to separate the methoxy compounds from the corresponding hydroxy compounds, the mixture benzol + glacial acetic acid proved successful as a solvent. The separation takes 20-80 minutes, according to the solvent. A summary is given of the phenolcarboxylic acids in grapes and vine leaves. Free salycilic acid and gentisic acid in grapes are only to be found in the later maturity stages. The main differences in the phenolcarboxylic pattern of the cultivars investigated are caused by the quantitative differences in syringic acid, ferulic acid and two other components which are not as yet identified.
