Evaluierung von Kartoffelgenotypen bezüglich Trockentoleranz

Zur Identifizierung und Validierung von Kandidatengenen für die Trockentoleranz in Kartoffeln wurden 40 S. tuberosum-Genotypen in einem Gefäßversuch unter einem Rain-out Shelter unter Kontroll- und Trockenstressbedingungen evaluiert. Je Genotyp wurden 24 In-vitro-Pflanzen in 5l Töpfen bis zum Beginn der Knollenbildung unter Kontrollbedingungen kultiviert. Danach wurden die Pflanzen Trockenstress (20 Tage kein Wasser) ausgesetzt bzw. als Kontrolle weiterhin optimal bewässert. Probenahmen von Blättern in den Stress- und Kontrollpflanzen ermöglichten die Bestimmung spezifischer Inhaltsstoffe zu sechs verschiedenen Zeitpunkten. Weiterhin wurden etwa 50 Kandidatengen-Primerpaare für eine Expressionsanalyse mittels Realtime RT-PCR entwickelt. Die nach Beendigung der Stressphase unter Kontrollbedingungen bis zur Ernte kultivierten Kartoffelgenotypen wurden hinsichtlich phänotypischer Merkmale evaluiert. Nach der Ernte erfolgte die Charakterisierung der Erträge, der Ertragsparameter sowie der Knolllenqualität. Der Versuch zeigte eine große Variabilität der ausgewählten Sorten bezüglich ihrer Reaktion auf den Trockenstress. Die Kombination der phänotypischen mit den genotypischen Daten wird zur Entwicklung molekularer Marker beitragen, welche eine gezielte Selektion trockentoleranter Kartoffelgenotypen ermöglichen.

Abstract
For the identification and validation of candidate genes for drought tolerance in potatoes, 40 S. tuberosum genotypes were evaluated in a pot experiment in a rainout shelter under control and drought stress conditions. Per genotype 24 in vitro plants were cultured in 5l pots under control conditions until the start of tuber formation. Thereafter, the plants were subjected to drought stress (20 days no water) or in case of the controls were optimally irrigated. Sampling of leaves in the stressed and control plants allowed the identification of specific components related to drought stress at six different times. Furthermore, about 30 primer pairs for candidate gene expression analysis using real time RT-PCR were developed. After termination of the stress phase the potato genotypes were cultured under control conditions until harvest and were evaluated with regard to phenotypic characteristics. After harvest, yield, yield parameters and tuber quality were characterized. The trial showed a great variation of the selected varieties with respect to their response to drought stress. The combination of the phenotypic and the genotypic data will contribute to the development of molecular markers which allow a targeted selection of drought-tolerant potato genotypes.
